Humanized Mouse Models
Despite numerous information obtained from mouse models, there still exist limitations to mouse models when applying to explore human biology. Several constituents of biological systems of mice, especially the immune system, are incongruent with those of humans. In addition, lots of pathogens and drugs are species-specific. The property and pathogenesis of immune responses reacted to pathogens that can only infect human cells may be distinctly different from those of murine infectious agents or murine-adapted human infectious agents. More appropriate mouse models,which can recapitulate human biological systems precisely, are urgently required for oncology research.
Immunodeficient mice implanted with functional human cells or tissues, so-called the “humanized mouse”, have gradually proved to have significant importance in the pre-clinical study of human disease. Since immunodeficient mice undergoing mutations in the IL2 receptor common gamma chain (IL2rgnull) have already been described in the early 21st century, researchers have been capable to engraft murine recipients with human hematopoietic stem cells that can develop functional human immune system in mice. These humanized mice can be engrafted with human tissues, for example, liver, skin, islets, and majority of solid/hematologic cancers.
Application
Humanized mice permit the establishment of human immune systems, including functional T and B cells, after engrafting human hematopoietic stem cells (HSC) in body. Humanized mice also assistant in heightening engraftment of primary human tumors and malignant progenitor cells, for the sake of in vivo investigation of pathogenesis and function. What's more, the application of humanized mouse based tumor model may ensure implementation of truly “personalized” anti-cancer therapies in the clinic.
