Molecular Mechanism of ActionThe assay relies on the enzymatic breakdown of the labeled substance Ac-VEID-pNA (N-Acetyl-Val-Glu-Ile-Asp-p-nitroanilide) by Caspase-6, which subsequently releases the pNA (p-nitroaniline) component. The absortion peak of the freed pNA is at 405 nm (εmM = 10.5) and its concentration can be quantified by measuring the absorption at 405 nm or by comparing it to a standard calibration curve prepared using the pNA Dye Standard. This can be accomplished using a microplate reader or spectrophotometer.
