Encoded by the fibroblast growth factor 2 (FGF2) gene, it functions via binding to fibroblast growth factor receptor (FGFR) proteins. FGF2, or basic FGF (bFGF), controls numerous fundamental biological processes such as cell proliferation and self-renewal and also functions as one of the most critical components of commercial iPSC medium. Not only in traditional 2D culture mode but also in 3D culture pathway forming embryonic bodies, FGF2 STAB in E8-based customized media maintains human iPSC growth in typical cell morphology (Fig. 2A). Besides, FGF2 STAB supplemented in StemFit Basic medium can fit reduced weekly feeding schedules without losing cell pluripotent compared with commercial mTeSR medium (Fig. 2B). Our cost-effective FGF2 also performs well in human iPSC culture.

Compared with negative control (N.C.), all human mesenchymal stem cells (MSCs) treated with corresponding FGF2 in same concentration show stronger CD73 expression (Fig. 3A). In a 7-day cell culture trial without medium refreshing, FGF2 STAB exhibits the highest proliferation promotion indicating its outstanding thermal stability, then FGF2 group and positive control (P.C.) group (Fig. 3B). In a 21-day normal cell culture assay, all FGF2 keep a fibroblast-like morphology of MSCs and high-level CD73 expression (Fig. 3C). Similar conclusion can be generated on normal human dermal fibroblast (NHDFs) following similar workflow, such as maintenance of activated ERK/MAPK pathway reflected by high phosphorylated ERK1/2 signals (Fig. 4A, 4C), and notable growth stimulation by FGF2 STAB (Fig. 4B).
