Introduction of CACNA1A

The Voltage-dependent P/Q-type calcium channel subunit alpha-1A (CACNA1A) is encoded by the CACNA1A gene and is located on chromosome 19. To date, most of the CACNA1A mutations detected in EA2 patients have resulted in a truncated CACNA1A protein, while missense mutations have led to a familial hemiplegic migraine.

Function of CACNA1A Membrane Protein

Voltage-dependent calcium channels mediate the entry of calcium ions into excitable cells and are also involved in a variety of calcium-dependent processes, including muscle contraction, hormone or neurotransmitter release, and gene expression. The calcium channel is a multi-subunit complex composed of α-1, β, α-2/δ, and γ subunits. Channel activity is guided by the pore-forming alpha-1 subunit, while other channel activities act as auxiliary subunits that regulate this activity. The unique properties of the calcium channel type are primarily related to the expression of various alpha-1 isoforms, alpha-1A, B, C, D, E and S. The CACNA1A gene encodes the alpha-1A subunit, which is primarily expressed in neuronal tissue.

    This article demonstrates that the IVS3-S4 and IVS5-SS1 linkers play an essential role in determining multiple biophysical and pharmacological properties of alpha1A-containing Ca2+ channels.

  2. Ophoff R.A., et al. Familial hemiplegic migraine and episodic ataxia type-2 are caused by mutations in the Ca2+ channel gene CACNL1A4. Cell. 1996, 87(3): 543-52. PubMed ID: 8898206

    Authors in this group find four different missense mutations in conserved functional domains. One mutation has occurred on two different haplotypes in unrelated FHM families. In EA-2, they found two mutations disrupting the reading frame. Thus, FHM and EA-2 can be considered as allelic channelopathies.

    This article shows that EA2 and SCA6 are the same disorder with high phenotypic variability, at least partly related to the number of repeats, and suggests that the small expansions may not be as stable as previously reported.

    This article focuses on the first point mutation of the CACNA1A gene. It not resulting in a proposed truncated protein. Furthermore, this mutation has been detected in a family member with mild clinical signs including only migraine.

    This article evaluates that the novel S218L mutation in the CACNA1A calcium channel subunit gene is involved in FHM and delayed fatal cerebral edema and coma after minor head trauma. This finding may have important implications for the understanding and treatment of this dramatic syndrome.
