What is the Complement Test?

Complement testing is a method used to assess the function of the immune system. The complement system is a complex protein network system consisting of more than 30 soluble proteins, membrane-bound proteins and complement receptors. Complement tests mainly include the following methods: Complement function or activity assay - to assess the overall function of the complement system. Complement factor concentration assay - to measure the amount of specific complement proteins. Complement activation product assay - to detect fragments or shear products produced by the activation of the complement system. Complement autoantibody assay - to examine the presence of autoantibodies against complement proteins. Complement gene analysis - to study gene variants associated with complement.

Case 1

Functional evaluation of complement activation through hemolytic assay techniques

To investigate complement activation initiated by antibodies under therapeutic hypothermia conditions, researchers performed functional hemolytic assays at these temperatures. Specifically, they tested antibody-sensitized sheep erythrocytes incubated in normal human serum and assessed hemolysis. In this CH50 assay, the degree of hemolysis varied notably with temperature changes.

What experimental methods can be applied to complement analysis in an experimental setting?

(a) In vitro experiments with human serum and blood. We can easily study the activation mechanism and intervention of complement activation in vitro with human serum. (b) Animal experiments. We can apply genetically defective or knockout animals, such as those with genetic defects in individual complement components. (c) Testing animal complement. Hemolysis assays can be designed for any species.

What types of samples can I submit for testing?

We can work with a variety of samples, including serum, plasma, whole blood, and tissue samples. Please prepare them according to our guidelines to ensure the best results.

What are the special requirements for sample collection and processing?

Complement assays require high quality samples. Blood samples should be separated from serum within 2 hours of collection and frozen quickly. Avoid repeated freezing and thawing to prevent decreased complement activity. Fresh serum may be required for some tests.

Can multiple complement components be tested at the same time?

Our custom complement multiplex assay services allow you to simultaneously assay two or more complement proteins, including complement C2, complement C4b, complement C5a, complement C5, complement C9, complement mannose-binding lectin (MBL), complement factor D, complement factor I, and complement factor P.
