Introduction | Complement factor I (CFI), also known as C3b/C4b inactivator, is a glycosylated protein in the complement system that encoded by the human CFI gene. Acting as plasma trypsin-like serine proteinase, CFI was first identified in 1966 as an enzyme involved in the physiological degradation of C3, a major plasma complement protein. CFI is a heterodimer consisting of a disulfide-linked heavy chain and light chain, which plays an essential role in regulating the immune response by controlling all complement pathways. It inhibits activations of all three complement pathways by cleaving and inactivating C4b and C3b to regulate the levels of C3 and C5 convertases. The diseases associated with CFI include Complement Factor I Deficiency, Recurrent Infections, Atypical Hemolytic Uremic Syndrome, and Macular Degeneration Age-Related 13. |
