| Introduction |
Complement component C3a (C3a) is an unglycosylated polypeptide released from the C3 convertase cleavage of complement component 3 (C3), the central protein of the three pathways of the complement cascade. C3a functions as an anaphylatoxin by binding to the C3a receptor (C3aR), mediating a series of inflammatory responses such as smooth muscle contraction, vascular permeability increase, histamine release, mast cell degranulation, and chemotaxis.
Similar to C5a, C3a is rapidly catalyzed by endogenous carboxypeptidase B through the removal of the C-terminal arginine to be an "inactivated" C3a desArg within few minutes. However, unlike C5a desArg, C3a desArg has no proinflammatory activity. C3a desArg is also known as acylation stimulating protein (ASP), which is named for marked stimulating action on triacylglycerol synthesis, glucose transport, and fatty acid uptake by binding to the C5L2 receptor in human adipocytes and fibroblasts.
As C3a desArg is the main active form of C5a in plasma and serum, the quantitation of C3a-desArg in samples is an indirect reflection or determination of the level of complement activation in the test samples. |
