What’s SP-A?

Surfactant protein A (SP-A) is a hydrophilic glycoprotein synthesized by alveolar type II epithelial cells. In addition to the lungs, it is also localized in extrapulmonary sites such as saliva, amniotic fluid, and semen. SP-A plays a role in many biological processes. During respiration, surfactants effectively reduce surface tension and protect the alveolar space from collapse. The SP-A protein is also one of the original agents of the innate immune system. As a lipid and carbohydrate recognition molecule, SP-A can recognize and bind to specific regions of pathogenic microorganisms, and then interact with phagocytes to regulate the release of various signaling molecules and initiate the process of innate immunity. Depending on the receptor it binds, SP-A can inhibit allergy, regulate mast cell proliferation, participate in cell polarization and exert inflammatory/anti-inflammatory effects. Purification of SP-A is the basis of research on surfactant proteins.

Here, we briefly describe the protocol for purifying natural SP-A from amniotic fluid by affinity chromatography. Similar procedures can be used with simple modifications for the purification of SP-A from other samples.
