NAA Introduction
Natural autoantibodies (NAA) are a kind of immunoglobulins that are spontaneously and constitutively secreted by the B1 type of lymphocytes in the absence of external antigen stimulation or immunization. Particularly belonging to IgM isotype, NAA play an important role not only in eliminating invaded pathogens but also in maintaining homeostasis of the immune system by prevention of autoimmune and inflammatory reactions. Low-serum titer, low-affinity to antigens, polyreactivity and connectivity are the main characteristics of NAA. In the early stage of some diseases, NAA are biologically amplified and hence may be measurable, making them promising biomarkers to discriminate the pathological changes from healthy tissue accurately. NAA detection and profiling present an alternative to clinical diagnosis with advantages of minimalist hardware, short time-frames, and an abundance of targets.

Various Applications of NAA
With the development of high throughput technologies and complementary advanced sequencing technologies, NAA are detectable in blood samples from patients or healthy individuals, which guarantees a wide range of clinical applications of NAA.

Application in IVIg
Widely used in a broad range of autoimmune and systemic inflammatory disorders, intravenous immunoglobulin (IVIg) has become a novel and efficient method in the clinic. IVIg is a therapeutic pool containing normal polyspecific human IgG obtained from large numbers of healthy donors, thus IVIg contains the wide spectrum of NAA and ‘immune’ antibodies expressed in normal human serum. IVIg has shown good curative effect with prolonged suppression of disease-related autoantibodies in patients with autoimmune diseases treated with IVIg.

Clinical Diagnosis
The profiling of the NAA repertoire with protein arrays might expedite the diagnosis and therapy of immune disorders, allowing not only the rapid diagnosis after the onset of the disease, but also the early detection of individuals susceptible to develop immune disorders. A large number of NAA targets have been discovered in recent years, which may be potential clinical indicators for disease diagnosis.

Cancer neoantigens can reflect tumorigenesis, but they are hardly detectable at the early stage, while NAA can bind to cell surface antigens on malignant cells, recognizing the cancerous cells, discriminating early cancer from healthy tissue, and retarding tumor development. So, NAA play an important role in immune surveillance against cancers.
