Aluminium Based Minerals
The majority of currently available vaccines contain a single adjuvant, “alum” (s mixture of aluminum phosphate and aluminum hydroxide), which was introduced by Alexander Glenny in 1926. Although aluminum salts have been in widespread use and have a good safety record, they are weak adjuvants for antibody induction and induce a T helper-2 (Th2) -skewed, rather than a T helper-1 (Th1) response, an attribute that has been implicated in the rapidly waning humoral responses to re-emergence of pertussis in the USA and elsewhere in the world. Thus, more effective adjuvants are required to realize the full potential of subunit vaccines.
Oil-Based Emulsions
Saponins, which are steroids or triterpene glycosides found in many wild and cultivated crops, are popular immune stimulants for inactivated vaccines. Saponins-based adjuvants have the unique ability to stimulate cellular immunity and enhance antibody production. Ginseng, Astragalus, notoginseng and other herbs have gained attention as candidates for plant derived saponins.
Bacterial Products
Bacterial products are considered as a good source of immune adjuvants because of their potent immunostimulatory capacity. Bacterial flagellin is a potent adjuvant to CD4+ T cells in vivo. Heat shock proteins (HSPs) are highly conserved proteins whose function may play a key role in the integration of innate immunity and adaptive immunity. [1]
