The surface-active protein SP-D, known as collectin, is a multifunctional, pattern-recognizing innate immune molecule. It is an important component of the body's defense system, participates in pathogen recognition and regulation of lung inflammatory response, and plays an inhibitory role in the pathological process of several lung infections. SP-D is predominantly present in type II pneumocytes, with the highest expression observed in alveoli. SP-D can also be detected and collected in extrapulmonary tissues, such as plasma, serum or amniotic fluid. Here, we briefly describe the protocol for purifying SP-D from amniotic fluid, which will serve as the basis for any SP-D-related research and provide a guarantee for the success of subsequent analysis experiments. This protocol is only suitable for the purification of SP-D in amniotic fluid samples, but can also be extended to other samples after simple modification.
