Monoclonal antibody (mAb) has become an important tool for the detection of specific substances in the field of biology and medicine. Hybridoma cell culture in vitro is an important way to prepare mAb. Traditional culture media need to be supplemented with serum, which will cause batch-to-batch differences, instability of the culture system, and risk of contamination. Creative Biolabs has considerable experience in the development and optimization of hybridoma. We have established a complete hybridoma suspension domestication system that can gradually transform the culture method from serum-containing media to serum-free media. Hybridoma can thus be cultured in suspension instead of semi-adherent style.

Overview of Hybridoma Culture

The most commonly used media formulations for hybridoma culture are Dulbecco's modified Eagle's medium (DMEM) and RPMI-1640, and these basal media are frequently supplemented with fetal bovine serum (FBS). Traditionally, tissue culture media have required serum supplementation to support the long-term culture of cells. FBS and other animal products, in general, are plagued by inconsistent product lots. Some lots of FBS may contain little contamination, and the nutrient or vitamin components may vary in different lots. All these inconsistencies have a significant impact on cells as well as their antibody production, which makes manufacturing difficult. More importantly, FBS contaminates antibodies which reduce the overall purity of the produced mAb. Serum-free media are highly effective for the culture of parental cells before fusion and even during the selection of hybridomas immediately after fusion. The replacement of the serum-containing medium with the serum-free medium provides great ethical and scientific advantages for further antibody purification.

Workflow of Hybridoma Suspension Domestication

Hybridoma suspension domestication is a method to adapt mAb-producing hybridoma cells from adherent culture to suspension culture, enhancing scalability and productivity. Initially, cells are transitioned from adherent to suspension culture conditions to promote growth in a free-floating environment. This is followed by gradual adjustment of culture parameters to optimize cell growth and antibody production.
