Foot and mouth disease (FMD) is caused by aphthovirus from Picornaviridae family. FMD virus has a positive-sense RNA genome that encodes a large polyprotein which also acts as the template for RNA replication. FMD is highly contagious of cloven-hooved animals such as cattle, swine, sheep, and goats with significant economic impact. The typical clinical sign caused by FMD is the occurrence of blisters on organization. The FMD viruses have seven immunologically distinct types, respectively are A, O, C, SAT1, SAT2, SAT3, and Asia1. The virus can be transmitted in many ways, including close-contact animal-to-animal spread, long-distance aerosol spread and fomites, or inanimate objects, typically fodder and motor vehicles. In a susceptible non-vaccinated population, morbidity could be as high as 100%. Vaccination is a well-established strategy for protecting susceptible animals against FMD. Emergency vaccination is the most powerful tactics and can play an important supporting role in the control of FMD outbreaks. Inactivated FMD vaccines
Inactivated whole FMD virus vaccines are the most widely used to control the spread of FMD virus. Currently available inactivated FMD antigen vaccines are applied intramuscularly to individual animals, lead to serotype and subtypespecific protection in 1-2 weeks. The killed virus preparations are pure, safe, and effective, where the virus has been subjected to a chemical treatment so it cannot reproduce in vaccinated animals.
DNA Vaccine against FMD
A novel DNA candidate vaccine was established that can aid in FMD control while providing differentiating infected from vaccinated animals (DIVA) capabilities. The negative antigenic marker virus candidates which derived by infectious cDNA technology, featuring the deletion of the leader coding region and one of the 3B proteins were constructed and evaluated. The technology of infectious cDNA clones encoding the marker viruses also contain unique restriction endonuclease sites flanking the capsid-coding region that allow for easy derivation of custom designed vaccine candidates. The attenuated and antigenically marked viruses can open a new window to virulent strains for FMD vaccine manufacturing.
Chimeric Vaccine against EMCV
O and SAT serotypes of FMD virus are relatively more thermolabile and their viral capsids readily dissociate into non-immunogenic pentameric subunits, which can compromise the effectiveness of FMD vaccines. Here we created the construction of a chimeric clone between the SAT2 and O serotypes, designed to have SAT2 antigenicity. Characterization of the chimeric virus showed growth kinetics equal to that of the wild-type SAT2 virus with better thermostability, attributable to changes in the VP4 structural protein. Sequence and structural analyses confirmed that no changes from SAT2 were present elsewhere in the capsid as a consequence of the VP4 changes. Following exposure to an elevated temperature, the thermostable SAT2-O1K chimera induced higher neutralizing-antibody titers in comparison to wild-type SAT2 virus. The chimeric O1K foot-and-mouth disease virus with SAT2 outer capsid could be an FMD vaccine candidate.
