Background of Anti-denatured DNA Antibodies

Anti-denatured DNA antibodies are a group of anti-nuclear antibodies (ANA) against denatured DNA strands such as double-stranded DNA (dsDNA) and single-stranded DNA (ssDNA). In 1957, they were found firstly in patients with SLE. There is a great deal of evidence suggests that the extracellular DNA is one cause of an immune response against dsDNA, and dead or dying cells are one major source of this extracellular DNA. Anti-denatured DNA antibodies can also be induced through infection via a mechanism known as molecular mimicry. The test of anti-denatured DNA antibodies is always using blood tests such as Enzyme-Linked Immunosorbent Assay (ELISA) and immunofluorescence (IF) in diagnostic laboratories. They are highly diagnostic of systemic lupus erythematosus (SLE) and are involved in the pathogenesis of lupus nephritis.

The Role of Anti-denatured DNA Antibodies in AIDS

Human immunodeficiency virus infection and acquired immune deficiency syndrome (HIV/AIDS) is an immune disease caused by infection with the human immunodeficiency virus (HIV). The increased incidence of HIV infection is demonstrated to be related to autoantibodies. The presence of anti-denatured DNA autoantibodies is significantly associated with lower CD4⁺ lymphocyte counts and increased mortality, which indicates a prognostic significance to detect autoantibodies in the context of HIV infection. Anti-denatured DNA antibodies were significantly more frequent in patients infected with HIV. Thus, the frequency and specificity of anti-denatured DNA autoantibodies in HIV-infected subjects can be regarded as a biomarker and an immunological hallmark of AIDS.
