Background of Anti-DNA Topoisomerase II

DNA topoisomerase II (TOP2), also termed as type II topoisomerase, is a kind of ubiquitous enzyme which plays a critical role in many fundamental DNA processes, especially in recombination and separation of daughter chromosomes, as well as chromosome structure. TOP2 alters DNA structure by generating a transient double-stranded DNA break, which is a fundamental action for all eukaryotic cells. TOP2 has two isoforms, TOP2α and TOP2β, which are differentially expressed in vertebrate species. Energy from ATP hydrolysis is required for TOP2 activity. Recent researches have expanded knowledge that the essential functions of TOP2 made a great contribution to prevent tumorigenesis. Anti-DNA topoisomerase II antibodies are autoantibodies directed against topoisomerase II that involve several diseases including autoimmune disorders, pulmonary diseases, and cancers.

The Role of Anti-DNA Topoisomerase II in Primary Liver Cancer

Hepatocellular carcinoma (HCC), a malignant tumor originating from the liver, is the main type of primary liver cancer that affects adults. It is the leading cause of cancer mortality in men worldwide, because the understanding of its pathogenesis is limited, and the current treatment effect is not satisfactory. TOP2α has been studied as target for cancer treatment in various cancer subtypes and as biomarker for predicting response. TOP2α plays an important role in preventing tumorigenesis, which has been researched as a sensitive and specific marker for proliferating cells and anticancer drugs for many years.

Current studies confirm that TOP2α overexpression is found in HCC, i.e., mRNA and protein levels of TOP2α are elevated. In addition, overexpression of TOP2α was detected in 72.5% of tumors, but not in matched non-tumors. Therefore, studies indicated that TOP2 autoantibody was a novel molecular that closely associated with transformation to primary liver cancer. More clinical tests and validation are needed to confirm that anti-DNA topoisomerase II can be developed as a potential marker for HCC diagnosis.
