Introduction of Direct Internalizing IntraMab Discovery

Direct internalizing intrabodies could target a variety of intracellular molecules to achieve therapeutic effects. They could be applied in the construction of immunotoxins, immunoliposomes, ADCs, and other targeted delivery architectures. As shown in Fig. 1, the scFv-based arginine fusion peptide and an immunoliposome molecule deliver the drug molecules into the cell to play a curative role.

Frequently Asked Questions

Q1: What's your strategy to generate the internalizing intrabody?

A1: To generate directly internalizing antibodies, our phage display library will be prepared for the target cell line. There are three stages for the phage internalizing process, binding to the cell surface, endocytosis, and internalizing within the cells. After the phage binding screen phase, non-specifically or weakly bound phage will be washed out. After the endocytosis screen phase, the phages binding to the cell surface antigens and in the extracellular matrix will be removed. Only the internalized phages will be recovered from within the cell and used for the next round of selection. After a few rounds of selection and enrichment, internalized phage clones will be further analyzed by a series of identification and characterization assays.

Q2: How to ensure that the intrabody was not internalized because of the phage?

A2: According to the screen design, the phage binding phase of the screen will be controlled to allow phage binding without internalization. The endocytosis phase of the screen will be controlled to allow the ligand-induced internalization of receptors and avoid phage degradation within the cells. All these efforts will help guarantee the get the direct internalizing intrabody will not be affected by the display vector phages.
