Cytoplasm Positioning
Based on our superior eukaryotic expression systems or customized cells, the screened intrabody sequence is primarily transcripted into messenger RNA (mRNA) first. Then the mRNA is released from the transcriptional machine, recognized and decoded by ribosome located in the cytoplasm by eukaryotic protein synthesis machine. After the transfer RNA (tRNA), free amino acids, elongation factors and other components belonging to the protein synthesis machine are involved in, the primary intrabody polypeptide is produced and is ready for the subsequent structure maturation and post-translational modifications (PTM), such as glycosylation which is extremely important for intrabody, acylation, prenylation, lipoylation, phosphorylation, carbonylation, biotinylation, ubiquitination, etc. Finally, the product intrabody without any N-terminal or C-terminal signal peptide targeting for other structure or organelles is residing in the cytoplasm.
Since the physiological conditions of cytoplasm such as pH and oxidative value, are sophisticatedly different from the extracellular environment, the intrabody residing in the cytoplasm needs to endure the harmful changes with undisturbed bioactivity and productivity. To address the technical bottleneck, our gifted scientists have developed multiple creative strategies to directly overcome or elaborately bypass it.
