Introduction of Intrabody

Distinguish from conventional antibodies which are designed to be secreted from the cell, intrabodies represent multiple formats of novel antibodies that have been developed and engineered for recognizing specific targets present in various organelles. With properly design and in frame fusion with intracellular trafficking/localization signals, specific intrabodies can be positioned on a series of subcellular locations, such as plasma membrane, cytoplasm, nucleus, endoplasmic reticulum (ER), plasma membrane and other subcellular locations. Currently, intrabodies have become one of the most cutting-edge hotspots, since they are feasible to be endowed with customized characteristics (high affinity via screening and optimization, specific cellular compartment retaining, fusion capacity, etc.) and the fact that most of the potential therapeutic targets are located intracellularly. Therefore, an increasing number of practical technologies of intrabody development have been optimized and employed for functional genomics, diagnostic, and therapeutic applications.
