What is the single domain antibody?

An intracellular functional single domain antibody (sdAb) is actually an antibody fragment consisting of a single monomeric variable domain of a heavy-chain Ab or of a common IgG with an MW of only 12-15 kDa (about 110 amino acids). The original discovery of sdAb is immunoglobulin new antigen receptor (IgNAR) of cartilaginous fishes (e.g. sharks). With the accumulation of research, the scientists have found heavy-chain Abs only in camelids (dromedaries, camels, llamas, and alpacas) among mammals.

Characteristics of single domain antibody

  • Specific as a regular antibody or even more robust.
  • More heat-resistance and stability in detergents and high concentration of urea.
  • More solubility in water originated from camelid and fish, owing to the CDR3, which forms an extended loop covering the lipophilic site that normally binds to a light chain.
  • Bulk production in bacteria or other expression systems due to the small size and single chain.

  • Chromobody (fusion of a fluorescent protein): recognize and trace targets in different compartments of living cells for novel functional studies.
  • GFP-nanotrap (couple of an anti-GFP sdAb to a monovalent matrix).
  • Crystallization chaperones: reduce the conformational heterogeneity by binding and stabilizing just a subset of conformational states as auxiliary protein, meanwhile, mask surfaces interfering.
  • Diagnostic biosensor: outperformance owing to the more dense couple on the biosensor surface, such as hPSA (human prostate-specific antigen) sensor.
