Background of NT5E

NT5E is also known as ecto-5'-nucleotidase (5'-NT) or CD73, which is ubiquitously expressed in most tissues. Generally, NT5E functions as a homodimer linked to the plasma membrane through a glycosylphosphatidylinositol lipid anchor to play roles in extracellular ATP metabolism. It can dephosphorylate AMP into adenosine rapidly after CD39 catalyzes ATP, ADP, and AMP. NT5E has been reported to participate in a variety of physiological responses including ischemic preconditioning, platelet function, hypoxia, vascular leak, and tissue injury. Especially, NT5E is up-regulated in various human cancers, including lung, colon, breast, pancreas, ovary and gastric cancer, indicating its potential as a possible target for cancer vaccines or immunotherapy.

The Role of Anti-NT5E Antibody in Gastric Cancer

Gastric cancer (GC) is a lethal disease with high mortality worldwide. The identification of novel biomarkers for carcinoma accurate diagnosis and novel therapeutic targets will improve the diagnosis, treatment, prognosis, and prevention of GC. (NAA)-based biomarkers attract particular interests because of the potential diagnostic and prognostic applications.

In human gastric cancer, the high expression of NT5E was correlated with oncobiology characteristic. It is documented that NT5E functions as a signal and adhesive molecule to regulate cell interaction with the extracellular matrix (ECM) components, such as laminin and fibronectin, and then mediate cancer invasive and metastatic properties. Importantly, NT5E can be recognized by autologous cytolytic T lymphocytes to elicit humoral and cellular immune responses, leading to the production of anti-NT5E autoantibodies. These autoantibodies show high-relevance with tumor differentiation, invasion depth, nodal status, metastasis, and tumor grade histology, as well as the overall survival rate and prognosis. Therefore, anti-NT5E or NT5E inhibitors might play a significant role in controlling tumor progression. At present, NT5E has been regarded as a highly significant prognostic marker for gastric cancer. Spontaneous anti-NT5E autoantibodies have been detected to help the early diagnosis.
