Chicken Astrovirus

Chicken astrovirus (CAstV) is a member of the recently confirmed avian astroviruses whose morphological structure and genomic condition are similar to those of other members of the family. The virus is round, non-enveloped icosahedron with a diameter of no more than 35 nm, the genome is single-stranded, positive-sense RNA, and the virions appear star-shaped under an electron microscope. CAstV can be transmitted through the fecal-oral route, and some strains can be passed from the mother of the spawning period to the chicks. The non-encapsulated nature of the virus makes it resistant to sterilization and cleaning.

CAstV is associated with broiler malabsorption disorders such as running stunting syndrome (RSS), enteritis, and uneven flock performance. The virus is able to infect chicks at the embryonic stage, and quantitative analysis in chickens indicates that the level of CAstV virus in the hatching chicks is higher and much higher than the viral load in the infected chickens after hatching. It is speculated that the age of the chicken is also a factor that affects the resistance of the chicken to the virus. In addition to the known fact that CAstV can cause enteritis, the virus can also infect areas outside the enteric tract including the liver and kidneys. Severely, India had a 40% mortality rate in broilers due to the serious kidney disease and visceral gout that caused by the viruses in 2012. Cases of "White Chicks" have been reported in countries such as North America, Brazil, and Poland. Chickens suffering from the disease have pale feathers and are weak, and generally, do not survive for a long time.

Immunity Against CAstV

Maintaining hygiene is an effective means of reducing CAstV infection. Providing enough CAstV antibodies to the eggs from the breeder hens is beneficial to prevent vertical transmission of the virus and delivery early protection for the chicks against the horizontal spread of the virus. There is some evidence that the age at which the chick is first infected with the virus is related to the progression of the disease caused by the virus, so early immune protection of the chick is necessary.

Development of Vaccines Against CAstV

The damage caused by CAstV infection to chickens can cause huge economic losses to the poultry farming industry. Moreover, the development of vaccines against CAstV has become necessary due to the virus's resistance to sterilization measures and insufficient effectiveness in maintaining hygiene against viral infections. The difficulty of CAstV's growth in cell culture makes the identification of virus-specific antigens relatively difficult. Therefore, the vaccines against CAstV currently mainly include inactivated virus vaccine and recombinant capsid protein vaccine. The results of indirect immunofluorescence assays, immunohistochemistry, virus neutralization assays, and ELISA assays indicate that capsid proteins of CAstV prepared using the recombinant baculovirus-insect expression system can induce chickens to produce certain levels of antibodies after inoculation with the protein, demonstrating immunogenicity of the protein. On the contrary, unpurified cell lysates prepared for commercial applications as vaccines may present problems with increased sensitivity for poultry to insects and extra immunopathology. In addition to the above issues, another challenge for the CAstV vaccine is the need to clarify the correlation between virus-specific antibody levels and protection effect produced by the vaccine.
