Ebola Vaccines

Ebola virus disease (EVD) is a rare and deadly disease in people and nonhuman primates which is caused by a virus of the family Filoviridae. The appearance of viruses is long filaments, sometimes branched or intertwined. The animal host of Ebola is widely believed to be the fruit bat and introduced into the human population through close contact with the blood, secretions, organs or other bodily fluids of infected animals and the incubation period is 2 to 21 days. Symptoms of EVD are the sudden onset of fever fatigue, muscle pain, headache and sore throat and followed by vomiting, diarrhea, rash, symptoms of impaired kidney and liver function, and in some cases, both internal and external bleeding.

EVD emerged at unprecedented epidemic levels in West Africa in 2014, caused fatality in 40 to 90 percent of cases and brought about a severe and often fatal viral hemorrhagic fever. In order to prevent EVD outbreak again, vaccination will be a key to controlling this. Several promising vaccine candidates have been shown to protect nonhuman primates against lethal infection.

DNA Ebola Vaccines

DNA vaccines are a very attractive choice for the development of vaccines against diseases causing sporadic outbreaks. The DNA vaccine encoding Ebola glycoprotein has been created which was encoded a transmembrane-deleted glycoprotein sequence from the Zaire and Gulu strain along with the nucleoprotein from the Zaire strain. The vaccine used the full length wildtype glycoprotein and contained two DNA plasmids in equal proportion. The candidate DNA Ebola vaccine has proved for an excellent safety profile.

Virus-Like Particles

Virus-like particles (VPL) is a protein-based approach where multiprotein structures are generated that mimic the organization and conformation of authentic native viruses but lack the viral genome, potentially yielding safer and cheaper vaccine candidates. The VLP-based Ebola vaccine encodes glycoprotein and nucleoprotein of Ebola virus was under development. Mixed with a saponin-based adjuvant, this vaccine possesses encouraging safety and immunogenicity specialties which give a huge potential to become a candidate for Ebola vaccines.

Recombinant Viral Vectors Vaccines

Viruses are designed to serve as antigen delivery platforms because of their ability to induce cellular immune responses as antigen are expressed and processed intracellularly from cells infected by the recombinant virus. The most advanced Ebola vaccines are viral vectors which include a recombinant replicating live vesicular stomatitis virus (VSV) Ebola vaccine and a chimpanzee adenovirus serotype 3 encoding Ebola glycoprotein from strains of EBOV and SUDV vaccine. These two vaccines are able to generate putatively protective antibody titers and the immune responses can be significantly enhanced. These advantages of recombinant viral vectors make them an excellent choice for Ebola diseases.
