Overview of POC

POCs are covalent constructs that link an oligonucleotide or its analog to a synthetic peptide sequence. Despite the high costs, POCs represent an increasingly successful strategy. POCs consistently exhibit strong biological effects in various applications. They involve modified peptides or proteins. POCs play a crucial role in the studies of the protein- and peptide-nucleic acid interaction. In addition, they can be used to improve cell-specific and intracellular delivery of antisense or antigene compounds. Notably, this technology is playing an increasingly important role in cancer immunotherapy, liquid biopsy, and single-cell analysis. Briefly, POCs can be used:

  • Increase the stability of the oligonucleotide
  • Template-enhanced POC coupling to larger molecules
  • As aptamers and nanomaterials
  • Intracellular delivery

Synthetic Strategies for POCs

There are two main approaches for synthesizing POCs, which are conceptually divided into on-resin synthesis of the entire hybrid molecule, and post-synthetic conjugation either in solution or on the solid phase, especially through click chemistries. The first method relies on the sequential coupling of amino acid monomers and nucleic acid monomers to generate diblock copolymers. The second method is more commonly used because it circumvents compatibility issues with solid-phase synthesis protocols and does not rely on foreign protecting group strategies. In this approach, each molecule is synthesized individually with unique reactive functional groups. The two moieties are then ligated directly followed by subsequent purification of the desired POC.
