Protein-Oligosaccharide Conjugation Services

Oligosaccharides are saccharide polymers containing two to ten monosaccharides. They usually exist in the form of glycans and play important roles in cell recognition and cell adhesion. The low immunogenicity of carbohydrate antigens makes it difficult to elicit an effective immune response. This problem can be solved by covalently linking it to an immunological protein. Protein-Oligosaccharide Conjugates Method

Protein-oligosaccharide conjugation through the use of dicarboxylic acid linkers is the simplest and most efficient method. During this process, free amino groups are introduced to the oligosaccharide antigen to facilitate coupling to the bifunctional linker upon reaction with the disuccinimidyl ester. The obtained monosuccinimidyl ester of the oligosaccharide antigen then reacts with the free amino groups of the carrier protein to finally complete the coupling of oligosaccharides to proteins. What's more, the oligosaccharides with amine-containing linkers can be conjugated to carrier proteins according to two-step squaric acid chemistry. SELDI-TOF-MS or MALDI-TOF-MS can be used to monitor this conjugation reaction. Moreover, a variety of cleavable linkers and non-cleavable linkers are all available.
