Introduction of Biotin
Biotin is a water-soluble vitamin also known as vitamin B7, vitamin H or coenzyme R. Avidin is a glycoprotein found in egg white, which is known for its high affinity for biotin. However, due to its high isoelectric point and carbohydrate content, avidin can bind non-specifically to other substances besides biotin. Streptavidin is a protein with similar biological properties to avidin, derived from Streptomyces avidinii. It does not contain sugar chains, thereby reducing non-specific binding issues associated with avidin. The strength of the biotin-streptavidin interaction has made it a valuable tool for assay design and specific targeting studies.

Application of Protein-Biotin Conjugates
The biotin-streptavidin system has the advantages of high specificity and strong binding capabilities, making it very versatile in a wide range of fields, including but not limited to:
Immunoassays
These include western blotting, immunoprecipitation, immunohistochemistry, etc. Biotin labeling significantly improves detection sensitivity compared to direct labeling with a detectable label.
Protein Purification
Utilizing a chromatographic column pre-fixed with streptavidin, biotin labeling can separate the target protein from the mixed solution.
Protein-Protein Interaction Assays
After immobilizing the proteins with the biotin-streptavidin system, the interaction between the two proteins can be analyzed by detection techniques such as surface plasmon resonance (SPR).
