Overview of PAL

In recent years, photoaffinity labeling (PAL) has gradually emerged as a promising technique to study protein properties and functions. PAL is a method of binding chemical tags to protein molecules with very high tag specificity. The basic principle of PAL is to introduce probes into the system under study and convert them into highly reactive intermediates capable of covalently binding to biomolecules by irradiation with near ultraviolet or visible light. In this process, probes are often analogs of complex molecules in which certain functional groups are replaced with photoreactive groups, such as azide, diaziridine, or benzophenone. Furthermore, in order to detect cross-linked products, the probe must contain a reporter group. Unlike chemical tags, photoaffinity tags are free to form covalent bonds between ligands and their specific binding proteins.

Applications of PAL

PAL occupies a crucial position in protein-ligand interactions. PAL can recognize unknown targets of ligands and play a crucial role in elucidating protein function and conformational changes, as well as identifying new binding sites in proteins. In addition, PAL has been used to analyze a range of small molecule interactions, including drugs, lipids, metabolites, and cofactors. Most importantly, target protein identification is a critical step in drug discovery and PAL can be used to identify binding proteins in complex mixtures of biomolecules. By this method, it is feasible to identify the amino acid residues located at these sites to obtain structural information of the ligand binding site. Therefore, PAL is an attractive strategy for drug development.
