Overview of Protein-Metal Chelates Conjugation

Proteins can be labeled with metal elements so that they can be used as imaging reporter molecules or for other applications. The steps of this technique are simple. The target protein first binds to a suitable chelating agent to form a stable complex. Specifically, the protein was first desalted into a suitable buffer, then a bifunctional chelating agent was added, followed by desalting the protein conjugate into an acidic buffer. The conjugate is finally labeled by adding the appropriate metal salt. There are different requirements for chelators. First, the chelator is able to react with proteins without adversely affecting their chelating properties. In addition, chelators form stable complexes with target metals that do not dissociate in their biological environment.

Applications

Some metallic elements have properties that make them attractive in a variety of imaging modalities. These properties mainly contain fluorescence, paramagnetism and radioactivity. In recent years, these properties have gradually become the focus of exploring the function of biomolecules in biological systems. Protein metal chelates obtained through a new series of bifunctional chelators will be suitable for biomedical applications. The final products of metal ion conjugates play important roles in radioimmunoassay, radioimmunotherapy, magnetic resonance imaging and photodynamic therapy. For example, these products can be used as in vivo probes in biological systems and in vitro probes in analytical systems such as radioimmunoassays. Furthermore, novel metal chelate protein conjugates are promising tools in tumor diagnostic imaging and tumor therapy.
