Intrabody Development through Prenylation
Prenylation is one of the most critical modifications for a proper physiological function of a natural protein, which is covalently attached to hydrophobic isoprene polymers (branched five-carbon hydrocarbon) at cysteine residues. In the natural bioprocesses, the most common isoprenoid groups are farnesyl (15-carbon) and geranylgeranyl (20-carbon) at cysteine residues via thioether linkages. As a reversible post-translational modification, dynamic prenylation is a determinant for specific protein-protein interactions and particularly critical for eukaryotic cell growth, differentiation, and morphology. Pathological deregulation of prenylation causes multiple diseases such as cancer. Moreover, the geranylgeraniol, farnesol, and dolichol eventually can be cycled to form cholesterol, which is related to many modern diseases.
