Background of Anti-Tau Antibodies

Tau proteins (also referred to as τ proteins) are a kind of neuronal proteins encoded by MAPT that promote the assembly and stability of microtubules. Tau proteins mainly distribute in neurons of the central nervous system (CNS) and are less common elsewhere, but are also expressed at undetectable levels in CNS astrocytes and oligodendrocytes. Defective tau proteins are or inappropriate stabilization microtubules can result in the pathologies and dementias of the nervous system, such as Alzheimer's disease (AD) and Parkinson's disease. In addition, tau proteins play a significant role in the establishment and maintenance of neuronal polarity since the hyper-phosphorylated tau proteins are the main component of neurofibrillary tangles. Anti-Tau antibodies are autoantibodies specifically against tau proteins, mainly IgM and IgG isotypes, produced in an autoimmune response and neurodegenerative process. Detection of these tau autoantibodies might provide a sight of AD diagnosis.

The Role of Anti-Tau Antibodies in Alzheimer's Disease

Alzheimer's disease (AD) is an age-related neurodegenerative disorder which recently has been suggested to be a kind of autoimmune disease. A variety of autoantibodies have been detected from patients with AD, among which anti-tau antibody is a low-level antibody. It has been demonstrated that autoantibodies of IgM isotype against phosphorylated-tau in the serum of AD patients increased significantly compared with the healthy controls. Consistent results have been obtained by the detection of anti-tau autoantibodies in cerebrospinal fluid of AD patients. Furthermore, the phosphorylated tau levels in AD patients were much higher in the saliva, an easily available body fluid sample relative to cerebrospinal fluid, than that in healthy human saliva. All these results indicate that salivary tau proteins and serum tau autoantibodies, especially anti-phosphorylated-tau IgM antibodies, can be used as a valuable biomarker in AD disease evaluation, more importantly, which also provide a direction for AD therapy.
