Tyrosinase Expression Assay

Background

Tyrosinase (also known as TYR, ATN, SHEP3, etc.), is the rate-limiting enzyme in the whole pathway of melanin production, primarily distributed in melanosomes of the melanocytes.

Tyrosinase is a copper-containing oxidase, which widely exists in animal, plant, and fungal species, activating the production of melanin and other pigments from tyrosine by oxidation. The expression level of tyrosinase is regulated by the upstream modulator microphthalmia-associated transcription factor (MITF), thus participating in multiple downstream pathways.

Tyrosinase Expression Assay in Pigmentation

In humans, melanin is the key enzyme of pigmentation in skin, eyes, and hair, protecting the tissues from damage caused by UV radiation. Its expression level and activity are directly associated with the synthesis of melanin. However, when the expression of tyrosinase is abnormally elevated, it induces hyperpigmentation disorders and raises risks of melanomas.

Considering the critical role in melanogenesis, tyrosinase has emerged as a hotspot for research and clinical projects, especially for skin diseases, as well as for commercial purposes in the cosmetic industry. Especially, inhibiting tyrosinase has emerged as one of the most attractive therapeutic strategies to deal with excessive pigmentation, and has inspired many related studies and trials. Accordingly, tyrosinase expression assay has become a requisite tool in those studies on pigmentation to evaluate the reaction of tyrosinase levels.
