Introduction to AAV Vectors
The attractive life cycle of AAV vector and its ability to infect a wide variety of mammal cells with stable expression have made it a perfect transfer vector in gene therapy. In the past few years, AAV vectors have been widely used for treating a number of diseases in animal models and clinical trials. The safety of AAV vectors has aroused much attention and the efficacy of AAV vectors still needs to be validated. As a result, a series of novel approaches have been developed for revealing the biological mechanism of AAV and designing more effective AAV vectors for gene therapy. For example, genome sequencing technology has been used for evaluating the integrity of AAV vectors and revealing the bio-distribution of the genome in the process of vector preparations. Meanwhile, long-term episomal persistence of AAV has also been assessed in a number of tissues, such as muscles, livers. The abundance of AAV circular intermediates has been analyzed by southern blot analysis with inverted terminal repeats (ITRs) and a green fluorescence protein (GFP) reporter.

Up to now, we also provide several unique services for analyzing the functions of AAV, including Recombinant AAV Genome Sequencing Service, AAV Biodistribution and Shedding Analysis Service, AAV Episomal Persistence Evaluation Service and AAV Integration Frequency Assessment Service. For instance, the integration frequency of recombinant AAV (rAAV) has been determined by using deep sequencing and PCR assays in our company. The data indicate that the integration of rAAV vectors should be related to many diseases in humans, such as chronic granulomatous disease. In addition, we have also conducted several projects for our clients to analyze the biodistribution of AAV-2, including shedding services and germline transmission investigation in mice models. The results have proved that an efficient and prolonged gene transfer of AAV-2 vectors can be achieved by following systemic administration.
