As an emerging field, the development of antibodies against cytoplasmic targets is gaining more attention. Many associated techniques have been developed to facilitate those projects. Antibodies or fragmented antibodies (e.g. scFv and single domain antibody VHH) can be raised against a cytoplasmic target by conventional methods such as hybridoma, phage display antibody library screening, and plasma cell sorting. However, different from antibodies against cell surface antigens, the evaluation of their in vitro efficacy requires their entering into the cytosol to reach the intracellular targets. Based on the assumption that smaller size increases the chance of membrane penetration, many anti-cytoplasmic protein antibodies are developed as antibody fragments. However, sometimes the small size does not present enough penetration and further modifications are still required to enhance the internalization capacity. Cell-Penetrating Peptides (CPPs)

CPPs are small peptides discovered in the late 1980s that contain unique sequence features. These sequence features render those peptides either highly charged or amphipathic. Proteins bearing CPPs often show higher cytosol entering potential, despite their source of origin. Creative Biolabs offers CPP-related modification services to promote the internalization capacity of either antibodies or proteins.

Lectin-Mediated Endocytosis

Some cell surface receptors bear lectin modules that recognize certain glycan formation and tend to form lectin-glycan complexes that will ultimately be internalized. We refer to this process as lectin-mediated endocytosis. With our advanced glycol-engineering platform, scientists at Creative Biolabs can help create antibodies bearing glycans via glycol-engineering and chemo-enzymatic glycan modification to take full advantage of the lectin-mediated endocytosis process.
