To help successfully introduce functionally active antibodies that target cytoplasmic proteins from outside the cell, Creative Biolabs has developed a series of intracellular delivery pathways based on the cationization process, by which we can achieve efficient delivery into numerous living cells without any genetic modification. The antibody cationization services contain two main strategies, protein cationization and cationic liposome-based systems. Our experienced scientists will design the most suitable approach for your project.
Creative Biolabs has developed perfect strategies to minimize side effects with more efficient intracellular delivery via protein cationization. This strategy involves chemical modification of proteins with diamines or polyamines, resulting in a positive net charge of the molecule, which can then be attached to the negatively charged cell surface by ionic charge interaction and subsequently be taken up by the cell. Ethylenediamine (EDC) and polyethylenimine (PEI) are the commonly used reagents, which can be chemically conjugated to a target antibody to generate artificially extensive and uniform cationization, endowing the antibody surface with an excess of overall positive charge.
Creative Biolabs possesses the first-rate cationic liposome-based delivery systems to provide customers with high-quality intracellular antibody delivery service, which includes TFA-DODAPL:DOPE, DOGS, DOPE plus CholCSper and DOPE plus SAINT-2 (N-methyl-4(dioleyl)methyl-pyridinium-chloride). This strategy is based on the fact that cationic lipids possess amphipathic nature with a hydrophobic core bridged by a linker group to a hydrophilic head group, which could form non-covalent complex with proteins, including antibodies. After attaching to negatively charged cell surfaces, the cationic lipids-protein complex undergoes endocytosis, and then fuses with but further escapes from the endosome to be released into the cytoplasm.
Superior features of our cationic liposome-based systems
As a novel lipid-mediated delivery system consisting of trifluoroacetylated lipopolyamine (TFA-DODAPL) and dioleoylphosphatidylethanolamine (DOPE), TFA-DODAPL:DOPE creates a protective vehicle for immediate delivery of functional proteins, such as antibodies, into living cells without significant toxicity.
Aiming at the intra-cytoplasmic delivery of anionic proteins, the DOGS-Cargo complex could buffer the acidity of endosomes and lead to endosome swelling and rupture via a ‘proton sponge’ mechanism, and subsequently, manifest the corresponding functions. For instance, the delivery of anti-β actin and anti-α tubulin IgGs leads to fiber depolymerization.
As a cationic surfactant consisting of cholesterol linked to carboxy-spermine by a cysteine, CholCSper combines with DOPE for an outstanding performance of cargo delivery.
The cationic amphiphilic lipid SAINT-2:DOPE manifests equal delivery efficiency for proteins including antibodies, as for nucleotides.
