TFA-DODAPL:DOPE based Antibody Cationization Service

The introduction of antibodies into the cytosol of living cells allows tracking at the molecular level and provides the opportunity to interfere with intracellular processes, which is valuable for both academic study and drug development. Introduction of TFA-DODAPL:DOPE

In nature, antibodies are extracellular proteins and the direct expression of antibody fragments in the cytoplasm of the target cell has proven to be much less reliable as specialized chaperones needed for antibody folding are lacking and the more reducing milieu impairs the formation of disulfide bonds. Therefore, it is highly desirable to have a broadly applicable method to introduce antibodies into living cells. With years of experience in antibody development and engineering, Creative Biolabs now provides this TFA-DODAPL:DOPE delivery system as a useful tool to help customers target antibodies to the cytoplasm. TFA-DODAPL:DOPE, also called BioPORTER, is a kind of cationic lipid formulation consisting of trifluoroacetylated lipopolyamine (TFA-DODAPL) and dioleoylphophatidylethanolamine (DOPE). Biochemically, the positively charged lipid formulation integrates non-covalently with the cargo proteins creating a protective vehicle for immediate delivery of functional proteins including antibodies into living cells without significant toxicity.

Although the efficiency of delivery varies primarily due to the properties (such as the net charge, hydrophobicity, lipophilicity, and conformation) of the molecule to be delivered and the targeted cell type, it still surpasses both microinjection and electroporation to be a powerful tool for functional genomics, cellular biology, and of great potential for therapeutic applications. Besides antibodies, a variety of different proteins can be delivered to the cytoplasm of numerous cell types by using this TFA-DODAPL:DOPE delivery system, including recombinant proteins, peptides, high and low molecular weight dextran sulfates, phycoerythrin, phycobiliproteins, albumin, beta-galactosidase, proteases, and caspases.

Compared to other protein delivery approaches using membrane transport peptides, our TFA-DODAPL:DOPE delivery system has two significant advantages:
