Description of SS

SS is one of the most common autoimmune diseases. It may exist as either a primary syndrome or as a secondary syndrome when associated with other autoimmune diseases, such as rheumatoid arthritis, systemic lupus erythematosus, systemic sclerosis, and primary biliary cirrhosis. The dominant picture is epithelitis caused by lymphocyte infiltration of exocrine glands, however, extra-glandular manifestations may affect above 50% of patients. Although the pathogenesis of SS is unknown, it is hypothesized that genetic predisposition with epigenetic modification underlies the disease and is further triggered by environmental factors. This is called a "mosaic of autoimmunity". At present, there is no common evidence-based intervention treatment, mainly symptomatic treatment. Therefore, further understanding of the pathophysiology of primary SS is crucial for finding new biomarkers and identifying new therapeutic targets.

SS Develop Pathway

The pathogenesis of primary SS may be a continuous, multi-step process leading to selective injury of the exocrine glands and to target organ dysfunction. Primary SS is the result of a complex interaction of multiple factors, including genetic and epigenetic control of immune homeostasis and gene expression, age and gender, and environmental damage. Newly discovered genetic associations in primary SS implicate disturbances in both the adaptive and the innate immune pathways.
