Feline leukemia virus (FeLV) is retroviral in the family Oncovirinae and is one of the most feared causes of a highly contagious and potentially fatal retroviral infection that weakens a cat's immune system. It is responsible for almost a third of cat cancer deaths and is transmitted mainly through saliva. The effects of FeLV are paradoxical, causing cytoproliferative and cytosuppressive disease. It manifests primarily through profound anemia, malignancies, and immunosuppression and infects domestic cats and other species of Felidae. There are four FeLV subgroups of clinical importance, namely FeLV-A, B, C, D, respectively.
Inactivated FeLV Vaccines
There is an inactivated FeLV vaccine available. It is a multiple viral antigen vaccine for vaccination of healthy cats 9 weeks of age or older as an aid in preventing persistent viremia, lymphoid tumors caused by FeLV, and diseases associated with FeLV infection. This vaccine is prepared by propagating FeLV, subgroups A, B, and C, in FeLV-transformed lymphoid cells and the viral antigens are chemically inactivated, combined with a sterile adjuvant to enhance the immune response. Clinical results have demonstrated that the vaccine has the ability to protect cats in high-risk environments where frequent or continuous contact with known leukemic cats occurred.
Combined FeLV Vaccines
Two combined vaccines are available to protect cats from FeLV infection. One is a modified live virus for protection against panleukopenia, calici, rhinotracheitis, chlamydia, and feline leukemia. Another is for protection against FeLV, rhinotracheitis, calici, panleukopenia viruses and as an aid in the reduction of the severity of disease due to feline Chlamydia psittaci. They are safe and efficient for cats and kittens.
Subunit BLV Vaccines
A subunit FeLV vaccine was developed from a molecularly cloned inactivated whole-FeLV. It was assessed for its ability to protect cats against homologous and heterologous virulent viral challenge. The results demonstrated that this vaccine prepared from a molecularly cloned subgroup A FeLV produces a high level of protective immunity against heterologous and homologous FeLV infection. Moreover, a subunit expression system was under development, as a means of vaccinating cats with both the env and gag proteins of FeLV which were expressed in a thymidine kinase-negative feline herpesvirus and a baculovirus. After vaccination, 100% protection against FeLV challenge was achieved using an immunization schedule which utilized both env and gag products delivered at both a mucosal and systemic site. The results show these strategies are very important for the protection of cats against FeLV.
DNA Vaccines
A strategy of DNA vaccine was designed due to the cell-mediated immunity is important in the control of FeLV infection. The vaccine consisted of two plasmids, one expressing the gag/pol genes and the other expressing the env gene of FeLV-A and the genetic adjuvants were plasmids encoding the feline cytokines interleukin-12 (IL-12), IL-18, or gamma interferon (IFN-γ). After vaccination, the vaccine consisting of FeLV DNA with the IL-12 and IL-18 genes conferred significant immunity, being safe to administer and conferring a degree of protection from transient, persistent, and latent FeLV infection.
