Simian immunodeficiency viruses (SIVs) are a large group of genetically related viruses belonging to the family Retroviridae, subfamily Orthoretrovirinae and genus Lentivirus. SIVs cause persistent infections in at least 45 species of African non-human primates and the natural hosts of SIV include many species of African monkeys and the chimpanzee. SIVs refer to a diverse group of viruses that vary considerably in pathogenesis and virulence in their natural host species or macaques. They are members of a genetically diverse group of lentiviruses isolated from African primates including two human immunodeficiency viruses (HIV-1 and HIV-2). SIV is transmitted through contact with infected body fluids such as blood.

Live Attenuated SIV Vaccines

The live attenuated SIV was constructed by deletion of the nef gene from simian immunodeficiency virus (SIV) strain SIVmac239. When inoculated in macaques with the vaccine, it offers substantial protection against a subsequent challenge with some SIV wild-type viruses. The results can lead to a conclusion that the attenuated growth and protection observed with the SIVΔnef vaccine strain does not require that the virus shift its characteristic site of replication, the CD4+ T lymphocyte. In fact, this immunodeficiency virus can replicate actively in CD4+ T cells prior to being contained by the host, at least in part by a strong killer cell response that is generated acutely in the infected lymph nodes.

Subunit Vaccines

The efficacy of nanoparticles (NPs) adjuvants was investigated in inducing protective immunity against simian immunodeficiency virus (SIV). These NPs were constructed by antigens adjuvanted with ligands specific for Toll-like receptor 4 (TLR4) and TLR7/8 encapsulated in poly (lactic-co-glycolic) acid (PLGA)-based nanoparticles. Rhesus macaques (RMs) were immunized with NPs containing TLR4 and TLR7/8 agonists mixed with soluble recombinant SIVmac239-derived envelope (Env) gp140 and Gag p55 or with virus-like particles (VLPs) containing SIVmac239 Env and Gag. NP-adjuvanted vaccines induced robust innate responses, antigen-specific antibody responses of a greater magnitude and persistence, and enhanced plasmablast responses compared to those achieved with alum-adjuvanted vaccines. Moreover, they induced antigen-specific, long-lived plasma cells (LLPCs), which persisted in the bone marrow for several months after vaccination. This strategy identified early blood transcriptional signatures that correlate with Env-specific antibody responses in vaginal secretions and protection against infection. The results demonstrate the adjuvanticity of the NP adjuvant in inducing persistent and protective antibody responses against SIV in RMs with implications for the design of vaccines against human immunodeficiency virus (HIV).
