Jaagsiekte sheep retrovirus (JSRV) is an exogenous betaretrovirus belonging to the family Retroviridae that is the causative agent of lung cancer in sheep known as ovine pulmonary adenocarcinoma (OPA). The disease is a slowly progressive, contagious viral infection and is spread predominantly through the respiratory route and may also infect lymphocytes and myeloid cells, in addition to the lung epithelia. JSRV envelope protein is oncogenic by itself which through the cytoplasmic domain of the transmembrane glycoprotein and some domains of the surface glycoprotein. Nowadays, OPA is a significant economic and animal welfare problem for the sheep industry in many countries.
Recombinant JSRV Vaccines
JSRV21 is a full-length JSRV provirus, possessing the hallmarks of integrated retroviral proviruses, which completely sequenced and showed open reading frames in agreement with the original strain of JSRV. In the vaccine design strategy, recombinant JSRV21 was produced in vitro by transient transfection of 293T cells with plasmid pCMV2JS21. A transient JSRV-specific cellular response was measured in the group of sheep immunized with this recombinant vaccine and a lymphoproliferative response to JSRV was observed in animals immunized, which shows that this vaccine has a consequence of some immunosuppressive activity of JSRV.
Subunit Vaccines
The subunit vaccine was constructed. Recombinant Gag proteins encompassing the JSRV-capsid (CA) and nucleocapsid (NC) domains of JSRV were produced in E. coli and injected into sheep. Besides, the JSRV capsid protein was also produced as a poly-histidine tagged protein by sub-cloning the CA-coding region of JSRV into pIVEX2.4a. JSRV-CA administered in CFA or IFA induced an antibody response and can successfully induce a JSRV-specific humoral response. These strategies of subunit vaccine can be the options in the future JSRV vaccine development.
DNA Vaccines
Another vaccination strategy that has been tried in sheep is DNA vaccination, followed or preceded by injection of protein antigens. This has been successfully employed in sheep against pathogens where antigen-specific humoral and cell-mediated responses were generated. For respiratory pathogens such as JSRV, mucosal immunity is likely to be important for protection. Plasmid pCI-CA was created by sub-cloning the region of JSRV21 encoding CA into the mammalian expression vector pCI using standard cloning techniques and checked by DNA sequencing. Cultured 293T cells were transfected with pCI-CA and CA protein expression was confirmed by immunoblotting and detection with an anti-CA rabbit polyclonal antiserum. After vaccination in sheep by intranasal inoculation, it has demonstrated that strong mucosal responses can be established by this DNA vaccine.
