Mouse Mammary Tumor Virus

Mouse mammary tumor virus (MMTV) is a type of retrovirus that is transmitted by milk. It is also the most important pathogen leading to breast tumor in mice and the vast majority of mouse mammary cancer is caused by the virus. Some mice endogenously carry MMTV, and the virus can also be transmitted vertically from the mother to the progeny via milk. MMTV is often integrated into the DNA in lymphocytes in milk in the form of a DNA provirus. After the virus is transported into the Peyer's patches, the host's macrophages are infected there, followed by infection of the lymphocytes. It has recently been discovered that MMTV is not a simple retrovirus in that it can encode a self-regulatory mRNA export protein, Rem, which has a similar function to the Rev protein of HIV. Like other retroviruses, MMTV has the ability to integrate its genetic information into host DNA, but when its proviral DNA is inserted into or near an oncogene, the expression of the oncogene may be activated. In turn, it causes cancer. Endogenous MMTV can react with hormones that regulate mammary growth and development as well as physiological processes such as breastfeeding. When the mouse enters puberty, the virus begins to express its mRNA.

Development of Vaccines against MMTV

More and more studies have found that there is a certain relationship between MMTV and human breast cancer, and the presence of MMTV is also detected in human breast tumors. Therefore, the study of MMTV vaccines has great implications for both the protection of mice and the study of vaccines for human breast cancer. At present, there are many directions for the research of MMTV vaccine, including inactivated whole virus vaccine and protein vaccine or polypeptide vaccine targeting gp52 protein or its epitope of the virus. In addition, in-depth research between MMTV and human breast cancer is also an important idea to design and prepare human breast cancer vaccine.
