Upon activation of the complement system through any pathway, enzyme complexes known as C3 convertases are formed, which cleave C3 into two fragments: C3a and C3b. The C3a fragment, composed of 75 amino acid residues, is released from complement C3 during system activation to stimulate the immune response. As the smaller fragment, C3a, akin to C5a which is subsequently generated, functions as a pro-inflammatory signaling molecule, known as an anaphylatoxin. Anaphylatoxins function as chemoattractants, drawing in and activating a range of inflammatory cells, such as mast cells and neutrophils.

C3a serves as a pivotal chemotactic mediator in the immune system, with the C3a/C3aR pathway playing dual roles—anti-inflammatory and pro-inflammatory—across various cells and diseases. By performing functional or activity assays of C3a, its function can be rigorously evaluated and quantified. Statistical analyses, such as presenting values as the mean ± standard error or mean ± standard deviation, are employed to determine the significance of differences using unpaired and paired t-tests. Spearman's correlation test is utilized to compare plasma concentrations of C3a.
