Overview of C5a Functional Test

Complement C5a

C5a is a pro-inflammatory peptide generated during the activation of the complement system. It is generated through the cleavage of complement component C5 by the enzyme C5 convertase. C5a acts by binding to specific receptors (C5aR and C5L2) on immune cells, triggering chemotaxis, enhancing vascular permeability, and promoting the release of pro-inflammatory cytokines. It plays a crucial role in immune responses by attracting neutrophils and macrophages to sites of infection or injury, contributing to inflammation, tissue damage, and the defense against pathogens. C5a is also involved in autoimmune diseases and other inflammatory conditions.

Complement C5a Functional Test

Method

The C5a functional test assesses the activity of the complement component C5a. Commonly used methods include C5a ELISA, which quantifies the concentration of C5a in serum or plasma, and chemiluminescence assays, which measure the activation of immune cells in response to C5a. The hemolysis assay involves testing C5a-induced red blood cell lysis. These methods help evaluate complement system activation and C5a's role in immune responses.

Analysis

Interpreting the C5a functional test involves analyzing C5a levels or activity in the sample. Elevated C5a concentrations may indicate excessive complement activation, suggesting inflammation, infection, or autoimmune disease. The test plays a crucial role in diagnosing conditions like sepsis, rheumatoid arthritis, and other immune disorders. It helps evaluate complement system functionality, guiding therapeutic interventions to manage inflammatory or immune-related conditions.
