Current diseases therapies utilizing the traditional small molecular agents would result in inadequate or adverse effects due to the lack of selectivity of cancer cells over normal cells, the rapid clearance of these water-soluble small molecular agents from the bloodstream, and the drug resistance commonly exhibited by tumor cells. CPP
CPPs are short peptides that promote cellular intake/uptake of a variety of molecules such as nanosize particles, small chemical molecules and large fragments of DNA. The "cargo" is related to the peptides either through chemical linkage via covalent bonds or through non-covalent interactions. CPPs typically have an amino acid composition that either contains a high relative abundance of positively charged amino acids or has sequences that contain an alternating pattern of polar/charged amino acids and non-polar, hydrophobic amino acids. A third class of CPPs is the hydrophobic peptides with only a polar residues and low net charge or have hydrophobic amino acid groups that are critical for cellular uptake.

Delivery System Based on CPP
It has been widely studied by researchers recently that CPPs can facilitate the intracellular delivery of various cargoes without causing any cellular damage. They are able to penetrate into a wide variety of mammalian cells independent of tissue or organism types. Furthermore, some CPPs can even promote targeted delivery to subcellular structure such as cell nucleus, mitochondria, lysosome and cytosol. Cargoes that have been delivered successfully by CPPs include small molecules, peptides, proteins, nucleic acids, quantum dots, polysaccharides, liposomes and nanoparticles.

The CPPs enter the cells by various mechanisms and thereby provide the possibility to transfer cargoes. The relatively short peptides composed of up to 40 amino acids have demonstrated to carry several times larger molecules without impact on their membrane penetration ability. These promising attributes render the increasing use of CPP in modern drug delivery systems.
