Targeting Spleen
The targeted delivery system has been exploited as a therapeutic tool in various treatments using the appropriate homing module, including antibody scaffold protein, peptides, and aptamers. Targeting spleen delivery is based on a unique set of marker molecules on endothelial surfaces within the spleen. The spleen is a lymphoid organ and is extensively involved in the various immunological functions of the body, such as removal of particulate matter, microorganisms, and abnormal red blood cells (RBC) from systemic circulation and elicitation of immune responses. The spleen has unique anatomical features including specialized microvasculature environment and slow blood circulation rate which presents immense opportunities for targeting drug delivery to the spleen. Homing modules such as peptides identified by phage display screening technique have been successfully used and promise to give good delivery of drug molecules or other agents to designated sites. Due to the difference in blood flow between spleen and liver, splenic delivery is inversely proportional to the hepatic uptake. Thus, it is desirable engineering of nanocarriers, which, upon intravenous administration, can avoid uptake by hepatic Kupffer cells to enhance splenic localization. With the progress of organ-targeting technology, many non-biological spleen-targeted drug delivery systems such as liposomes, polymeric nanoparticles, solid lipid nanoparticles (SLN), and inorganic nanoparticles have been exploited for spleen targeting and enhanced splenic uptake.

Advantages of Delivery to the Spleen
There are several advantages of spleen-targeted delivery. For instance, the enhanced splenic concentration of active agents could provide therapeutic benefits in spleen resident infections and hematological disorders including malaria, hairy cell leukemia and autoimmune hemolytic anemia. Besides, the enhanced concentration of radiopharmaceuticals in the spleen may improve visualization of the spleen, which could provide benefits in the diagnosis of splenic disorders.
