Targeting Cytosol
Targeted delivery of substances into cells has long been hampered by the plasma membrane, which is an efficient barrier to exclude most molecules that are not actively imported by living cells. The discovery of CPP (cell penetrating peptide) opened up new possibilities for the translocation of larger molecules through the plasma membrane. Armed with innovative and diversified module delivery systems, a good approach can be accomplished by combining the module targeting cell surface with CPP targeting cytosol together. The strategy of Cytosolic Targeting
Delivery of macromolecular drugs or smaller bioactive molecules is an important field in medical research. CPPs are unique tools to gain access to the cell interior and deliver a bioactive cargo into the cytosol. CPPs may have implications in biomedical sciences by improving the delivery of a wide variety of drugs through the membrane barrier. CPPs initially bind to negatively charged head groups of lipids or proteins in the plasma membrane followed by their internalization. Advances in recent years imply uptake mechanisms that are predominantly endocytotic. Normally, any alteration introduced in the peptide sequence will have a direct effect on the intracellular localization profiles.

A novel, cationic cysteine-rich CPP, cytosol localizing peptide-1 (CyLoP-1), has been developed and it exhibits distinguished diffused cytosolic distribution along with endosomal uptake at low micromolar concentrations. The efficient intracellular delivery and the distribution into the entire cytosol make it an appropriate candidate for cytosolic delivery of small sizes cargoes.
