Lumpy Skin Disease
Lumpy skin disease (LSD) is an economically damaging infectious bovine disease caused by the lumpy skin disease virus (LSDV), which is a member of the genus Capripoxvirus of the Poxviridae family. The virus is transmitted mainly mechanically by blood-feeding flying insects, such as Stomoxys spp. and Aedes spp. This disease is characterized by fever, lumps and edema, mucosal damage and lymphadenitis on the surface of the internal organs, which are occasionally fatal. It has spread to most African countries, Middle Eastern countries and recently to Europe. Due to its potential for rapid spread and great economic impact, LSD is listed as an OIE-notifiable disease.

Vaccines for Lumpy Skin Disease
Vaccination plays a key role in controlling the spread of disease. Vaccines currently used to protect cattle from LSD are primarily live attenuated vaccines based on attenuated strains of wild isolates passaged by cell culture. There are three licensed vaccines for bovine dermatosis (LSD): lumpy skin disease virus (LSDV) Neethling vaccine, Kenyan sheep and goat pox (KSGP) O-180 strain vaccines and Gorgan goat pox (GTP) vaccine.
LSDV Neethling Vaccine
The Neethling strain (Onderstepoort Biological Products, South Africa) developed through multiple passages of cell culture does not cause systemic infection or serious disease in cattle. It produces a local response to a granuloma of 1-2 cm in diameter at the inoculation site in 50% of the vaccinated animals, and the temporary decrease in milk production in dairy cattle has a negative impact on the use of this vaccine. In addition, in recent decades in Africa and the Middle East, the use of Neethling vaccines has successfully controlled the spread of LSDV with high levels of vaccine coverage.
KSGP O-180 Vaccine
The Kenyan sheep and goat pox (KSGP) O-180 strain vaccines (KSGP O-180) was isolated from a sheep during the same epizootics as the KSGP O-240 strain and has previously been used successfully as a vaccine against SPPV, GTPV, and LSDV. The KSGP O-180 vaccine reduces the severity of LSD infection and reduces susceptibility to LSD virus but increases the infectivity of vaccinated infected cattle. In general, vaccines are not effective in protecting cattle from LSD, either through direct clinical protection or by reducing transmission. Therefore, there is an urgent need to develop LSD vaccine development to control LSD in popular countries and prevent it from spreading to other countries.
Gorgan GTP Vaccine
The live attenuated Gorgan goat pox vaccine is produced to use against goat pox virus (GTPV) and lumpy skin disease virus in the Middle East. Compared to the LSD Neethling and KSGP O-180 vaccines, the GTP vaccine elicits a stronger immune response in the herd. A higher Delayed Type Hypersensitivity (DTH) response is observed in vaccinated cattle, indicating that the Gorgan GTP vaccine is more immunogenic. The close correlation between GTPV genomic sequence and LSDV may be the reason why GTP vaccine shows better protection against LSDV. The Gorgan GTP strain vaccine does effectively protect all calves from the challenge of virulent LSDV wild-type strains, suggesting that the Gorgan GTP strain may be the preferred candidate vaccine for controlling LSD infection.
