C4 Purification Protocol

Derived from the human leukocyte antigen (HLA) system, complement component 4 (C4) is an important protein in the complement system that, together with other components, performs many key functions in immunity, tolerance and autoimmunity. In order to better carry out research on the complement system, Creative Biolabs has developed many methods for the purification of various components. Here, we describe the C4 purification protocol for our clients all over the world.

For C4 protein isolation, the monoclonal antibody affinity column has been developed to distinguish C4-A from C4-B. Serum containing both isoforms was applied to an L003-coupled Sepharose column, then washed and eluted via a buffered pH gradient. Two clear peaks arose; SDS-PAGE analysis, based on the slower migration of the C4-A α-chain, confirmed their identities. Immunotyping with anti-Rodgers and anti-Chido sera verified the separation, with only minimal cross-contamination. Processing serum from individuals expressing a single allele yielded pure proteins. Recovery was no less than 80%, about 1-3 mg per 10 mL serum, obtaining C4-A3, A4, A6, and C4-Bl/B2.
