Introduction of Glycoprotein Quantification

Glycosylation is an integral part of proteomics, which is a very critical modification of therapeutic proteins, known to significantly modulate yield, bioactivity, solubility, stability against proteolysis, immunogenicity, and clearance rate from circulation. The glycan groups of glycoprotein mediate interactions of all cells, including reproductive cells, and regulate fundamental biological, chemical, and physical processes including evolution. The preferred source for detecting hepatocellular carcinoma (HCC) is serum samples, which contain multiple biomarkers, such as alpha-fetoprotein (AFP), but AFP levels are correlated with tumor size. To improve the specificity and sensitivity of HCC screening, it is urgent to develop new serum biomarkers. Researchers developed a quantitative glycoproteomics method, using lectin affinity chromatography to first enrich glycoproteins in serum samples, and then quantitatively analyze glycoproteins in HCC and control serum samples by mass spectrometry-based proteomics methods to discover and develop new serum biomarkers. The results showed that a total of 2280 proteins were identified in serum samples of HCC patients. In addition to AFP, the researchers found three differentially expressed glycoproteins by Western blotting, namely FOS-like antigen 2 (FOSL2), fibrinogen gamma chain (FGG), and α-1,6-mannosyl glycoprotein 6-β-N-acetylglucosaminyltransferase B (MGAT5B), which were all upregulated in HCC serum samples. It was inferred that these differentially expressed glycoproteins might serve as potential biomarkers for HCC.
