Abnormal glycosylation is an important hallmark of cancer. In this study, the authors used serum samples from healthy individuals as a control group to study the O-glycosylation characteristics of apolipoprotein C-III (apoC-III) in the serum of colorectal cancer (CRC) patients, thereby evaluating the differences in O-glycosylation of apoC-III in CRC patients. The authors directly analyzed the O-glycosylation of apoC-III protein in serum samples using a simple positive ion mode matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) technique. This analysis process did not require specific protein separation, was relatively simple to operate, and achieved high-throughput analysis. The results showed that compared with the control group, the O-glycosylation of apoC-III in the serum samples of CRC patients changed significantly, especially the sialylation level was significantly increased.
