NK cells serve as immune cells and can kill tumor cytotoxic cells. The authors developed a glycoengineering strategy to modify NK cells to give them the targeting ability to recognize B-cell lymphoma. CD22 is a selective target for B-cell lymphoma, the authors successfully introduced 9-O-modified sialic acid-based CD22 ligands onto the surface of NK cells through metabolic glycoengineering and tested their activity in vitro and in mouse tumor models. The results showed that glycoengineered NK cells had a significant ability to bind and kill CD22+ lymphoma cells. This metabolic glycoengineering method is effective and convenient, which is a new anti-cancer immunotherapy strategy.
