  • Sample preparation

We grind the biomass sample into powder, then use solvents such as ethyl acetate and n-hexane to extract terpene in the sample and concentrate it by rotary evaporation to obtain a concentrated terpene sample.

  • Chromatographic separation

We use gas chromatography to separate various terpenoid compounds in the sample. We choose the HP-5MS capillary column with a good separation effect as the chromatographic column, set a low initial temperature of the chromatographic column (for example, 50°C), and then heat it to a higher temperature at a certain rate. The separation of terpene components in the sample is achieved by gradient elution.

  • Mass spectrometry analysis

We use the internal standard method or external standard method to analyze the terpene components in biomass samples through a mass spectrometry detector. The internal standard method requires the addition of internal standard substances of known concentration to the sample, while the external standard method uses standard substances of known concentration to establish a standard curve. We identify various terpene components in the GC-MS chromatogram, such as pinene, linalool, limonene, β-myrcene, α-terpineol, etc. We compare the ratio of the peak area of terpene in the sample to be tested with the corresponding peak area in the internal standard or standard curve, calculate the content of terpene, and generate the final analysis report.

Terpenes are hydrocarbons that are classified according to the number of units or functional groups. Terpenes give biomass cannabis its unique odor and pharmacological properties. Currently, methods for quantifying terpenes in cannabis require large amounts of biomass samples, long extraction times, and long chromatographic elution times, which are not suitable for analysis in a high-throughput environment. Therefore, the authors developed a rapid GC-MS method to achieve high-throughput analysis of terpenes in biomass cannabis. They optimized the sampling technique, chromatographic columns, and solvents. The results showed that on and DB17 columns, the resolution of each sample peak was high, and all compounds could be eluted within a run time of 30 minutes. In addition, the peak shape of compounds extracted with hexane was good. Therefore, the authors used hexane to extract terpenes from cannabis and performed GC-MS analysis using dodecane as an internal standard. The results showed that a total of 48 terpenoids were detected in biomass cannabis. Moreover, the authors performed methodological validation for 22 of these terpenoids, including parameters such as detection limit, quantification limit, linearity, precision, and accuracy. The authors believe that this GC-MS analysis method is suitable for high-throughput analysis of biomass cannabis.
